Shedding Light on Possible Individualized Treatment Strategies for Lung Cancer
on 30. Oct 2008 in cancer.

The National Human Genome Research Institute (NHGRI) funded a large consortim of medical centers to undertake the most comprehensive effort focused on understanding the genetic changes involved in lung adenocarcinoma. This form of lung cancer is the most common form of lung cancer and the number one cause of deaths from cancer. The average 5-year survival rate for this disease is currently roughly 15 percent, but earlier detection methods would increase that rate.

One outcome of this study has been the indentifyication of areas of research for new, more individualized approaches for detecting and treating this form of cancer. NHGRI Acting Director Alan E. Guttmacher, M.D., comments, “By harnessing the power of genomic research, this pioneering work has painted the clearest and most complete portrait yet of lung cancer’s molecular complexities. This big picture perspective will help to focus our research vision and speed our efforts to develop new strategies for disarming this common and devastating disease.”

Major Health Risks for the Obese
on 15. Oct 2008 in heart disease, apnea, joints, cancer, Health, Obesity, Obesity, weight loss.

There are over one billion people overweight people worldwide and one out of three children in America are overweight. Noted as an epidemic, increase in weight size is quickly becoming an issue of focus. Weight loss can be a hard thing to overcome, but many have found the motivation to do so and have limited their health risks due to being overweight or obese. Shedding excess weight will allow one to have many benefits in life that include virtually eliminating disastrous medical conditions.

Some of these health concerns are diabetes, heart disease, stroke, cancers, sleep apnea and arthritis. These medical conditions cause great harm to the body and many are life-long effects or even killers.

Type 2 diabetes may develop from being overweight. The body after time can no longer control the blood sugar that is normal regulated. If untreated, diabetes can cause other serious conditions such as heart attacks, poor circulation or even death.

High blood pressure is more likely to be at an elevated level if a person is overweight. High blood pressure can lead to heart attacks or strokes, the leading cause for death in men and women in America. Sudden death is often possible without advance warnings.

Being overweight has shown to actually cause some cancers. Cancers caused by being obese are colon, ovary, gallbladder, rectal, prostate or breast cancers. The more excess body weight a person contains, the greater the increased risk for developing these cancers regardless of cancer in one’s family history.

Sleep Apnea is a condition in which a person stops breathing during sleep and rest. Heavy snoring can be a signal pointing towards a sleep apnea condition. Those who have excessive fat stores around the chin and neck areas due to being overweight, can actually have this fat interfere with proper breathing or even halt the breathing process. Losing weight is proven to resolve this condition in the majority of patients, but if allowed to continue, it can cause heart attacks, heart failure and sudden death from lack of air to the body.

Being overweight places more tension on fragile joints leading to pain and disability. Painful lower back pain or aching joints are often a sign of arthritis. Osteoarthritis is especially apparent in the obese. To reduce strain and stress on the joints, weight loss is a must in order to allow the joints to work and feel at ease.

The conditions abound for those that are overweight. Other conditions caused or has a much higher rate of occurring in overweight people include: an enlarged heart, pulmonary embolism, cysts developing in the ovaries, gastro esophageal reflux, fatty liver disease, hernias, depression, cellulitus, erectile dysfunction, urinary incontinence, chronic renal failure, gout and disease of the gallbladder. There are many more factors affected by being excessively heavy that involve weight loss to overcome or avoid. Losing weight definitely shows time shared with loved ones is limited when a person is overweight or obese and that should be the best reason for losing weight.

Sirtuin therapeutics for diseases of aging in Nature Reviews Drug Discovery
on 01. Oct 2008 in diseases of aging such as Type 2 Diabetes, mitochondrial disorders, sirtuin research, inflammation, Breakthrough Digest Medical News, heart disease, Breakthrough Digest Medical News, cancer.

Sirtris, a GSK company focused on discovering and developing small molecule drugs to treat diseases of aging such as Type 2 Diabetes, announced today that it published a new review article on the growing body of sirtuin research and its potential to treat diseases of aging such as Type 2 Diabetes, mitochondrial disorders, inflammation, cancer, and heart disease. Entitled “SIRTUINS – Novel Therapeutic Targets to Treat Age-Associated Diseases,” the review appears in today’s issue of the journal Nature Reviews Drug Discovery.

Sirtuins are a family of enzymes which target genes that control aging. There are a total of seven sirtuin enzymes that exist in mammals. SIRT1 is the founding member of this class of enzymes and is currently the most studied of the group. When activated, SIRT1 appears to mimic some of the positive health effects seen in calorie-restricted animals.

“We are excited to be at the forefront of this research, which continues to intensify as we see positive results from early human clinical studies,” said Christoph Westphal, M.D., Ph.D., Chief Executive Officer of Sirtris, a GSK company. “The body of clinical data supporting the role of SIRT1 activation as a viable mechanism for treating a broad range of diseases of metabolism and aging is growing.”

Peter Elliott, Ph.D., Senior Vice President of Development, said, “At Sirtris, we are targeting the genes which control the aging process with the potential to treat diseases of aging such as diabetes, neurodegeneration, cancer, and inflammation.”

This review highlights the molecular mechanism of action of sirtuins with a view towards diseases where SIRT1 activation shows therapeutic promise. Such diseases of aging include Type 2 Diabetes and mitochondrial disorders, cardiovascular disease, cancer, neurodegeneration and inflammatory diseases. One of the first small molecules discovered to exhibit the positive effects of calorie restriction is resveratrol, a natural ingredient found in red wine. Since then, Sirtris has developed its own proprietary formulation of resveratrol, SRT501. Early clinical studies in patients with Type 2 Diabetes indicate that SRT501 may lower glucose and improve insulin sensitivity. Sirtris also has new chemical entities (NCEs) which are structurally unrelated to and one-thousand times more potent than resveratrol. Sirtris’ first new chemical entity is currently being evaluated for safety and tolerability in a Phase 1a study in humans.

“We are beginning to understand more about activation of other enzymes in the sirtuin family, SIRT2-7, and we’re encouraged by early data that indicates a role for other sirtuins in multiple therapeutic areas,” continued Dr. Westphal.

Anti-tumor therapy guided by endoscopic ultrasound may allow doctors to fight cancer more safely and effectively

The chairman of EU󋈈 today announced that investigational research on a therapeutic technique that will allow physicians to directly inject malignant tumors with cancer fighting agents from inside the body will be presented at the 16th International Symposium of Endoscopic Ultrasonography (EUS2008) in San Francisco on September 12-13. This technique, which uses a flexible gastrointestinal endoscope with a miniature ultrasound transducer on the tip to guide a small needle directly into a tumor, could prove to be a safer and more effective approach to administering chemotherapy since it allows doctors to deliver therapy right to the tumor and avoid damaging normal surrounding tissues. Injecting drugs directly into the cancer using endoscopic ultrasound (EUS) in combination with systemic chemotherapy to kill cancer cells that have spread may prove to be a more effective approach to some cancers.

EUS combines endoscopy and ultrasound in order to obtain the most accurate, high resolution images and information about the digestive tract and the surrounding tissue and organs. A more advanced form of EUS, called curvilinear EUS, allows doctors to operate within the lumen of the gut while at the same time detect, biopsy and treat lesions and tumors that lie outside the intestinal wall. This technique is particularly useful in patients with pancreatic, esophageal and rectal cancer.

“Curvilinear endosonography will likely become the dominant technology within the field of EUS,” said co-chairman of EUS2008, Robert Hawes, M.D., Professor of Medicine and Peter Cotton Chair for Endoscopic Innovation at the Medical University of South Carolina. “The potential for accurate diagnosis using ultrasound-guided biopsy, precise staging with high resolution ultrasound images and then the enormous opportunity for new therapies with the curvilinear endoscope is why we are focusing this meeting on the use of this instrument alone.”

Used in conjunction with real time imaging, EUS can help physicians to detect blood flow in blood vessels in and around tumors as well as detect and biopsy tumors and lymph nodes as small as 3-5 mm. This allows doctors to avoid puncturing blood vessels when sampling tissue, get the most accurate view of the cancer and know exactly what stage a cancer is in for optimal therapy for treatment. This could save cancer patients with late stage disease from going through unnecessary surgery. EUS may also play a role in the future of minimally invasive surgery (MIS). A new paradigm in MIS is called natural orifice translumenal endoscopic surgery (NOTES®). This entails using the stomach as a window to the abdominal cavity rather than the skin. EUS could play an important role in helping surgeons gain safe access to the abdominal cavity as part of NOTES.

EUS 2008 will be devoted to teaching current applications of curvilinear endoscopic ultrasonography in order to encourage endosonographers and gastroenterologists to become proficient in these procedures, enhance their techniques and increase collaboration with oncology surgeons. This meeting has a rich tradition dating back to the first meeting held in Stockholm, Sweden in 1982. The meeting has evolved as technology has changed and improved along with the exponential growth of endosonographers around the world.
